New AI-Designed Drug Targets Mesothelioma’s Hippo Pathway

The Hippo signaling pathway has been identified as a key driver of mesothelioma tumor development. When working properly, it’s a biological control system that tells cells when to stop growing, but when it breaks down, cells instead receive a constant “keep growing” signal. Insilico Medicine, an AI-driven biotechnology company, has developed a novel drug targeting this key pathway involved in mesothelioma tumor growth and will present a report on the first-in-human Phase 1 trial results for ISM6331 at the upcoming European Society for Medical Oncology Congress in Madrid in October 2026.

Scientists have known for years that blocking the Hippo pathway could be an effective way to fight mesothelioma, but designing a drug small enough to enter cells and block the final switch in the pathway’s faulty signaling chain has been extremely difficult.  Insilico Medicine used its proprietary generative AI platform, called Chemistry42, to design ISM6331. According to the company’s representatives, the drug is designed to jam the “keep growing” message before it can reach the cells’ control centers.

The company has been engaged in a Phase 1 global multicenter clinical trial to evaluate ISM6331’s safety, including how well mesothelioma patients tolerate it, how the drug moves through the body, and early signs of whether it is effectively fighting tumor growth. First-in-human results will be presented in a Rapid Oral presentation at ESMO Congress on October 25, 2026, in Madrid.

“Patients with advanced mesothelioma and other Hippo-driven solid tumors face limited therapeutic options,” said Halle Zhang, Vice President of Clinical Development at Insilico Medicine. “These preliminary clinical findings provide early encouragement as we evaluate ISM6331’s safety and initial antitumor activity.”

ISM6331 joins Orion Pharma’s ODM-212—another TEAD inhibitor that recently received Orphan Drug Designation from both the FDA and European Commission for mesothelioma—in targeting the same Hippo pathway, suggesting that multiple promising drugs attacking this mesothelioma vulnerability may eventually give oncologists new treatment choices for patients who have exhausted standard options.
